Magnesium NCA in Horses

Is it possible to an NCA on magnesium levels?

There were 6 horses who underwent each of the 3 treatments in a Latin square.
A blood sample was taken just prior to the horses being fed a bolus of magnesium aspartate. Samples were collected after 5 minutes of dosing.

The issues I have in this study is, after the bolus was fed, the horses were fed hay which they

would have consumed over several hours. They were fed again after the 8 hour sample (480 min) sample was taken. There is a constant influx of Mg and how it can be analyzed?

The information I have is the Total Mg consumed per day was 33 mg/kg BW, 39 mg/kg BW and 58 mg/kg BW.

Hi Chandramouli, NCA is predicated on a single dose event either naive or at steady state. Your experiment suggests that you have a bolus IV followed by continuous ingestion of hay, that you infer would have noticeable amounts of Mg; I would be interested to see the data as to whether this ingestion was significant as compared to the bolus.

Do you have a predose reading of Mg, is it less than 5% of Cmax? maybe for practical purposes you can disregard the hay intake and just perform a NCA simple IV bolus.

Otherwise I think you are looking at making custom model to describe this data and it’s going to be tricky as you have no real way of knowing the oral dose or rate of intake.
