Hi, can I get someone to verify my project is set up correctly to obtain an estimate for “F” from IV and SC data? I am following the examples guide but the example doesn’t have the F in the model. I’m assuming if bioavailabilty for IV =1, then bioavailabilty for SC = F*1 in the model. But my initial estimates are quite different from my NCA results. for example my NCA suggests F = 0.88 V (from IV) = 48 L/kg CL (from IV) = L/hr/kg this is a small peptide so I’m not surprised NCA doesn’t match up so closely but I’m surprisde by the orders of magnitude they are off by. Can someone double check my logic that my model is set up correctly for an absolute bioavailability model? thanks Elliot

ok, thanks…you confirmed I was doing that IVSC correctly in terms of setting up the model. and yes, I noticed a units error. my next follow-on question, is I have both repeat dosing and single dosing (different animals) in the same file. I am not seeing the doses being fit and the predicted plots (eg popDV, IPRED vs IVAR) make it seem like a straight line is being fit across all the trough values leading up to the full profile in the repeat dose group. I saw a separate post where Serge posted a model with single and repeat dosing and the predictive plots had clear profiles fitted for each dose so I’m concerned I’m not accounting fo the repeat dosing in the model Can someone look at this model attached to confirm its being set up correctly? thanks Elliot [file name=Cyno_SC.phxproj size=702663]Certara | Drug Development Solutions (686 KB)
