Code For Enzyme Inactivation

Hi All,

I was hoping to find a coding example of an indirect response model that accounts for enzyme inactivation instead of a reversible mechanism (i.e. use of Kinact instead of IC50’s). I have estiamtes of Kin and Kout, which i presume would represent the normal dynamics of enzyme turnover.

Many thanks

Kirk Bertelsen

Dear Kirk

Not sure it will help you but can we look at the reference below around equation 18-19.

Best Regards and good luck

Hi Serge,

Thank you for the response, and indeed, these are the models I am interested in. My issue is this:

I have an in vitro derived estimate of the inactivation rate constant for a compound/enzyme system (i.e. Kinact, and which I understand what this describes), but am having some difficulty understanding its relevance/translation to the cell killing constants (i.e. k (k = −ln SFt/AUC(0–t))) that are commonly used in this type of model…any comment on your part?

Also, do you have any recommendations on case examples to help people with developing skills with the phoenix modeling language? I am working through the reference guide, but am interested in seeing the actual coding language for more diverse examples from what is listed within the guided.

Again, many thanks for your inputs,

Kirk Bertelsen

Dear Kirk

I am teaching this week. Come back to me next week and send me the mathematical equations you would like to code and I may be able to help you. My expertise is more on the math/stat/code part, not biological one.

Best

Serge
