Phoeni NLME-IOV

Hi Could you please tell me how to arrange the data sets for modeling IOV in NLME, as i am getting the same results when i am using different data set for Occasion 1 and Occasion 2. I feel i am doing mistake in arranging the data sets are while selecting covariate labeling in NLME.

Can you perhaps post a project you’re having problems with as an example might be easier to explain with. Simon.

Hi Simon Thanks for your quick reply, please find attached data set which am trying to do IOV. Thanks Sundar

Dear Sundar I made an example based on your data set to have iov implanted. I changed the data set to add reset function as at each occasion the time is reset to 0. I added mdv to make sure the program does not use the true 0 values that sometime are problematic when fitting. I assumed 1 compartment model with tlag just as an example. I defined period as the occasion. Look at all this and let me know if you need further help. It is just an example, I did not optimize that data set. I guess you had problems because of the reset function that requires uncheck the sort button and then in input select reset. I used 4 as arbitrary number but that matches NONMEM assumption of washout in evid. BEST SERGE [file name=iov_example.phxproj size=1121948]Certara | Drug Development Solutions (1.07 MB)

Dear Serge Thanks once again for your swift response. Could you please share the password to my mail id to open the IOV folder. mail id - sundaramurugesan@lupinpharma.com Thanks Sundar

Dear colleague You should not have any password problem. I guess it is your internet server. Anyway, go to Box and you will be able to download the project I made for you based on your data set. best Regards Serge

Let me know if you can access to that site and download the project. I tried to send you a message but not sure you got it. best Serge Box

Hi Serge Great!!.. Thanks a lot for sending the example results. Now i understood the problem, however the number which you selected in reset column is based on NONMEM Trans number?. Just i need a confirmation on that. Thank you once again. Sundar

In nonmem, if you use evid=4, it reset all the compartments to 0. In Phoenix NLME you can use whatever number you want as long as you write the sane number in your data set and in the reset input option. I wrote from 4 to 4 in the interface. It means that the program will reset the compartments to 0 when he sees in the reset column a number between 4 and 4 which is of course 4. We have just more flexibility and the number is arbitrary (you just need to match the numbers). best Serge

Sundar, This has been discussed on the forum a few times and it’s an artefact of the browser typically IE, e.g.; Certara | Drug Development Solutions if you’re using IE in particular as your browser it “helpfully” renames the PHXPROJ file to Zip (since it is in fact a compressed file). Just download the file then rename the extension back to xxx.PHXPROJ, there is no password protection on Serge’s file when opened in Phoenix. Simon.
