RSABE in PHX

Hi All, Greetings! I am a beignner in learning statistical software , one of my initial assignment is to run RSABE in phoenix and match it with the SAS results ( which is provided by my Boss). s2wr=0.14962 theta=0.79669 y=-0.1192 boundy=-0.063407 boundx=0.12176 sWR=0.3868 Estimate=0.15342755 StdErr=0.10665716 LowerCL=0.04208706 UpperCL=0.34894216 pointest=1.16582 x=0.012164 critbound=0.015946 I have followed the outlined workflow given in BEBAC forum and got CVwr & sWR, pointest value correctly for Cmax. Estimate is coming 2.54 in my case in the intermediate ilat step which is a problem. Kindly check the attachment of my project ( Raw data was there I have run NCA on that and extracted Cmax results seperately and again prepared the data, by using this data I have followed the workflow) and please tell me where I am lacking. Thanks & Regards. [file name=Cmax_data.xlsx size=11313]Certara | Drug Development Solutions (11 KB)

Hi Anu - is it possible you can attach your project as it will be easier to see the steps of your workflow there. thanks, Simon.

Hi Simon, Thanks for your reply. Yes I am attaching a pdf file here from which I have worked on PHX . The data set has been attached in the previous post. Thanks & Regards [file name=Replicate_USFDA_by_winnonlin.pdf size=707228]Certara | Drug Development Solutions

Hi Anu, THX, I know my own white paper. Please upload your [size=4][color=#FF0000]project-file[/color][/size]!

Hi Helmut, I thought have to attach the workflow( i.e your white papers) from where I have try doing my project. Kindly check I have attached the PHX file. Thanks & Regards [file name=Cmax.phxproj size=1790908]Certara | Drug Development Solutions

Hi Anu, a few problems: You have to adapt the filters R1 and R2 according to your coding of sequences. R1 should select the first administration of R (irrespective of the period) and R2 the second. Therefore, the filters should be: Form=‘R’ and ((Seq=‘RRT’ and Period=1) or (Seq=‘TRR’ and Period=2) or (Seq=‘RTR’ and Period=1)) Form=‘R’ and ((Seq=‘RRT’ and Period=2) or (Seq=‘TRR’ and Period=3) or (Seq=‘RTR’ and Period=3)) In ‘Pointest x and boundx’ you forgot the first transformation ‘pointest = e^x’, the filter should be Replace where [Dependent] = ‘ilat’ with ‘dlat’ That’s the reason why in the later ‘Merge 2’ you got two rows instead of one. The Transformation in ‘Dij Complete rows’ should be ‘lat1-lat2’; lat1 mapped to lat1r and lat2 to lat2r In ‘Intermediate dlat’ remove the ‘Estimate’ statement In the last Data Wizard add a Custom Transformation, Column Name ‘Assessment’, Formula: if(sWR<0.294, ‘sWr<0.294: use unscaled ABE’, if((pointest>=0.8) & (pointest<=1.25) & (critbound<=0), ‘RSABE’, ‘not RSABE’)) Generally: If you filter for something select the respective column, not the entire dataset (Filter Specification > Search Area > Column) I got: Preliminary (based on IBE/PBE): pointest = 1.1658233 CVwr = 40.17 Assessment: ‘PE within 0.8-1.25 and CV≥30%: try RSABE’ … and matching all of your Boss’ results in SAS: s2wr = 0.14961626 theta = 0.79668871 y = -0.11919759 boundy = -0.063406802 boundx = 0.12176036 sWR = 0.38680261 Estimate = 0.15342755 StdErr = 0.10665716 LowerCL = -0.042086669 UpperCL = 0.34894177 pointest = 1.1658233 x = 0.012164264 boundx = 0.12176036 s2wr = 0.068254493 critbound = 0.015946007 CVwr = 40.17 Assessment: ‘not RSABE’ (because of the positive critbound) [file name=AnuCmax.phxproj size=1841217]Certara | Drug Development Solutions
