Simulating PKPD model using custom coding

Hi, I’m trying to simulate Testosterone in a more complex mechanistic PKPD model. For example, the PK model has two absorption depots with different abs rates and the PD model has three separate compartments (LH pool, LH plasma, and Testosterone). I was able to use custom coding in NLME to get decent PKPD parameter estimates. However, now I want to simulate Testosterone levels at different doses. I fixed all PKPD parameters (pls see attached code). The sim file I used has the timepoints and dosing amounts I want to use. Pls note this is a simulation using naive-pooled method. The result for TstObs keeps coming out as -1 and simulated DV also comes out as -1. Is there a step that I missed ? Lisa, sorry I’ve been away for a couple of weeks and it seems this was overlooked - if you haven’t fixed this already then I think for a simulation you should be looking at Tst for the simulated values, but it would be much easier to trouble shoot this problem with the project so we can check your other settings.
