Fitting zero order release invivo data

Hello

Iam trying to fit microsphere formulation invivo data using different approaches

a) naive pooled model (naive NLME trial 4)

b) classical 2 comp model

c) transit compartment model (naive trial 1 &2)

but the issue is iam not able to capture the zero order absorption phase observed in the invivo PCT data

I have uploaded the invitro dissolution data

Can anyone help me in this iam attaching the PHX PRJ file for review purpose

Harish, I took a look at your project and used graphical mode to first try adding a zero order to the Absorption compartment but I think the fit was no better than your trial4 fit, both visually and by AIC.

I did also think I could try removing Ka from the model as the confidence was not good in this parameter’s estimation but still not much better in the last model I tried. Anyway take a look and see what you prefer.
