PK/efficacy modeling in Phoenix WNL

I’mm just wondering whether anyone has tried building PK/efficacy models in PML. I’m thinking of the work of Simeoni & Rocchetti on chemotherapies, or Jumbe et al (J PKPD, 2010) on T-DM1. I’ve built similar models in Classic WNL, but would like to get them going in PML too. In terms of specifics, I seem to be having trouble fitting against a control group (in which there is no dosing). The model appears to run, but there’s no output in the Simulation table (or anywhere else). Has anyone else tried this? Thanks, -Frank

Frank - on the simulation question. I assume you have requested a number of simulated points and/or a table ?

Hi Simon, Thanks for your suggestion. Yes, I did request points/timespan as you suggested. In fact, the problem turned out to be that I hadn’t yet figured out how to set the initial conditions on the tumor volume. However, since posting my original message, I’ve learned that the ‘sequence’ statement is how you do that, and it works beautifully. My next problem is how to link the PK to the PD in this model, using PML… Is there a concise way to specify multiple dosing regiments in PML, and is there any way at all to specify that each dose group gets a different dose? Or do I have to resort to those tedious tables of time/dose/group? Thanks for your input… -Frank
