We are looking to calculate the bioavailability of a prodrug as compared to the parent drug. We have calculated the NCA parameters for each individually (parent drug IV, parent drug PO, prodrug PO), but I’m not sure how to calculate the bioavailability of the prodrug using the parent drug information. They are different molecular weights and purities, so I’m not sure if that is in consideration. I also don’t know how to calculate bioavailability in WinNonlin in general for IV/PO group comparisons, so if there is a guide or tutorial for that, that would be really helpful! Usually I just do the calculation outside WinNonlin but if I can do it in WinNonlin, that would save me some steps.
I would recommend using the Data wizard and/or the Ratios tool
