MUTIPLE DOSE SIMULATION WITH PHOENIX MODEL

I have fitted single dose plasma concentrations following at oral dose of 6000000 ng in WinNonlin to get the fitted parameters (V/F=6382223.3; K01= 0.84428 and K10=0.301164) to the one cpt model.

In order to explore the capabilities of the NEW Phoenix model approach using graphics I tried to simulate the plasma concentrations following two doses using the above parameters except for the second dose I used K01=4 instead of =0.84428.

I cannot get this model to run. At an earlier date I had help from Simon and at that time he did get it to run. Let me attach the file. I am wondering whether there is a bug in the program or if I have altered the file inadvertently.

Hi Angus I opened the project and looked at the Warnings tab saved from your run and it seems there is something awry with your configuration as the message below indicates it can’t run the model engine.

If I execute your project file as I received it, it executes with no problem, so I think you need to work out what has changed since you last ran it OK. If you can’t resolve it yourself, I would suggest you contact Support (very easy now the Certara site uses the same login details/ password).

You need at add CObs in your template data set and map CObs with CObs in the main setting.

Thank you Serge: I downloaded your corrected file and tried to run it. It did not run. I do not see anything wrong with the model. Simon is suggesting a configuration problem, since he got the model to run without modification when I sent it to him. As he suggested I have contacted support.

Very strange. What version are you running?1.3 I guess. You could open my project? If so, can you send me back again the copied version that did not work. I want to see if nothing changed when copying.

If still does not work, then apparently it is nota model issue but something else not model related.

Serge: this morning I failed when I tried to run your file. Later on this morning I did some routine fitting- simulation in the original WinNonlin models (not Phoenix) and they worked fine. When I saw your reply I downloaded your Phoenix model file once again and tried to run it. I expected it to fail again: I ran it again and it worked! I have made no changes to your model at any time. I am thinking that there was a setting on my computer switched back to what it should be when I did the WinNonlin work. I have been failing for hours on this trying Phoenix models I have archived and run at an earlier date. I have not rebooted my computer today, since I tried that yesterday , but it did not help. I do pot understand it. I will try some of the other Phoenix models to see if they are running.

Serge: the model you sent to me is working. None of the others I have got working in the past are working. Please can you check the one attached. Can you tell me if it runs right away without you altering it?

If you change it and get it to work please can you tell me exactly step by step what you did to make it work. Either it is something very simple that I do not understand properly or I have a configuration problem here. The example I attach is to simulate 5 doses of drug.

Simon remarked that the other file I sent tom him worked right away.

SERGE: your model you sent to me originally from Simon runs well repeatedly from the Diagram view. By that I mean it runs over and over again. However; if you click on the Phoenix model box in the Diagram view to open it up and run the model from there then the model ceases to run.

I ran the model you sent as is and it ran by right away.

When I look at your warning nd errors I see

Access to the path ‘C:\Users\Angus\AppData\Roaming\Pharsight\Phoenix\PhoenixRemoteSettings.xml’ is denied.

It seems to be related uniquely to your computer but this should go to Phoenix technical support I guess as I have no issue from my location.

Serge/Simon: I have downloaded the new update to Phoenix: now I have run two of the problematic phoenix models without any problem.
